Labeling Disk Partitions using format command

I have a disk with free space that I want to use. I need to add it to slice 5 which is currently undefined. The other slices have active mounted file systems. Once I set up the size for slice 5, I need to label the disk.

Can I do a disk label with the active file systems mounted?
